How do you isolate lipoprotein

I would like to isolate LDL, and HDL and subject them to gradient electrophresis for particle size determination.I am just wonder if there is any one who has done this before. I have human plasma and serum samples. According to some paper that i read, the protocol involves adjustment of the density before ultracentrfugation with Kbr.However,, i do not know how to adjust density of plasma before fractionation. I really appreciate your help. If you could please be detailed as much as you can.

According to the attached reference, you can adjust the density of the plasma for centrifugation by the addition of 1.33g/ml of KBr.

In the paper which I think you're referring to here it gives the target density as 1.30g/ml, suggesting that the plasma itself must have a low to zero density. Although the attached paper references this group for the method, it provides a bit more practical detail.

Dear Martin, thank you very much for your response. I will go through the referred paper. - Rothman

No problem. Just for completeness according to The Physics Factbook blood plasma has a density of 1.025kg/l (or 1.025g/ml). - Martin Fitzpatrick
